What is trigonelline?
Trigonelline is a natural plant compound found in coffee beans and fenugreek, among other things, and is also present in your own body. It is a precursor to NAD+: a building block that the body can incorporate into its NAD+ supply. This places it in the same category as nicotinamide riboside (NR) and NMN, but it is the newest of the bunch.
What does the research say about trigonelline?
Attention to trigonelline received a significant boost from a study in Nature Metabolism (2024), by Nestle Research and the National University of Singapore. The researchers showed that trigonelline is incorporated into the NAD+ supply and increases NAD+ levels, and that lower levels in the blood are associated with muscle weakness (sarcopenia). In models, more trigonelline was associated with increased mitochondrial activity and better preservation of muscle function during aging.

Trigonelline and autophagy: the link explained
A term you hear more and more often surrounding trigonelline is autophagy. In simple terms: autophagy is your cells' cleanup and recycling system. Damaged or worn-out parts are broken down and reused, a kind of internal spring cleaning that is important for cells that age healthily.
What does trigonelline have to do with this? The link runs via NAD+. Trigonelline increases your NAD+, and NAD+ is the fuel for sirtuins, your cellular maintenance enzymes. Together with the energy sensor AMPK, these temper the mTOR signal, and it's precisely this push that triggers autophagy. In short: more NAD+ means more fuel for the system that keeps your cells clean and in shape.
To be fair, we stick to this: that this mechanism is correct is well-substantiated in preclinical research (cells and animals). However, it has not yet been definitively proven that trigonelline also measurably boosts autophagy in humans. It is a logical, promising link, and precisely for that reason interesting, but we present it as what it is: a strong mechanism, not a proven promise.
Trigonelline is promising, but early
